Os TAMs emergem como alvos atractivos para estratégias terapêuticas destinadas a reprogramar o seu fenótipo protumoral para uma atividade antitumoral eficaz. As evidências sugerem que estes TAMs promovem ativamente todos os aspectos da iniciação, crescimento e desenvolvimento do tumor. Assim, os tumores não devem ser tratados apenas como agregados de células cancerígenas, mas como estruturas separadas. O rácio alterado entre Foxp3+ Tregs e células Th17 desempenha um papel importante na patogénese das doenças relacionadas com o sistema imunitário. Apresentamos uma visão geral dos mecanismos responsáveis pelo recrutamento de TAMs e destacamos os papéis dos TAMs na regulação da angiogénese tumoral, invasão, metástases, imunossupressão e resistência aos quimioterápicos. Procurámos também descrever a interação entre as células Th17 e outras células imunes no microambiente tumoral, e avaliámos as potenciais actividades antitumorigénicas e pró-tumorigénicas das células Th17 e das citocinas que lhes estão associadas. Heutzutage gewinnt die Immuntherapie bei der Behandlung von Krebs immer mehr an Bedeutung. TAMs stellen attraktive Ziele für therapeutische Strategien dar, die darauf abzielen, ihren Protumor-Phänotyp in eine wirksame Antitumor-Aktivität umzuprogrammieren. Es gibt Hinweise darauf, dass diese TAMs alle Aspekte der Tumorentstehung, des Wachstums und der Entwicklung aktiv fördern. Daher sollten Tumore nicht nur als Aggregate von Krebszellen, sondern als eigenständige Strukturen behandelt werden. Das veränderte Verhältnis zwischen Foxp3+ Tregs und Th17-Zellen spielt eine wichtige Rolle in der Pathogenese immunologischer Erkrankungen. Wir geben einen Überblick über die Mechanismen, die für die Rekrutierung von TAMs verantwortlich sind, und beleuchten die Rolle von TAMs bei der Regulierung von Tumorangiogenese, Invasion, Metastasierung, Immunsuppression und Chemotherapieresistenz. Nowadays, immunity treatment is becoming more and more important in the treatment of cancer. TAMs emerge as attractive targets for therapeutic strategies aimed at reprogramming their protumor phenotype into an effective antitumor activity. Evidence suggests that these TAMs actively promote all aspects of tumor initiation, growth, and development. Thus, tumors should not be treated as only aggregates of cancer cells but as separate structures. The altered ratio between Foxp3+ Tregs and Th17 cells plays an important role in the pathogenesis of immune-related diseases. We present an overview of mechanisms responsible for TAM recruitment and highlight the roles of TAMs in the regulation of tumor angiogenesis, invasion, metastasis, immunosuppression, and chemotherapeutic resistance. We also tried to describe the interplay between Th17 cells and other immune cells in the tumor microenvironment, and we assess both the potential antitumorigenic and pro-tumorigenic activities of Th17 cells and their associated cytokines. ¿We believe this knowledge will be of great interest to researchers who is working on TAM-targeting therapy as a promising novel strategy for an indirect cancer therapy.
