The book focuses on protein allostery in drug discovery. Allosteric regulation, 'the second secret of life', fine-tunes virtually most biological processes and controls physiological activities. Allostery can both cause human diseases and contribute to development of new therapeutics. Allosteric drugs exhibit unparalleled advantages compared to conventional orthosteric drugs, rendering the development of allosteric modulators as an appealing strategy to improve selectivity and pharmacodynamic properties in drug leads. The Series delineates the immense significance of protein allostery-as demonstrated by recent advances in the repertoires of the concept, its mechanistic mechanisms, and networks, characteristics of allosteric proteins, modulators, and sites, development of computational and experimental methods to predict allosteric sites, small-molecule allosteric modulators of protein kinases and G-protein coupled receptors, engineering allostery, and the underlying role of allostery in precise medicine. Comprehensive understanding of protein allostery is expected to guide the rational design of allosteric drugs for the treatment of human diseases. The book would be useful for scientists and students in the field of protein science and Pharmacology etc.

This volume is the first-ever complete treatise on polymyxins and presents the most comprehensive and up-to-date reviews of all major research and clinical topics from chemistry, microbiology, pharmacology, clinical use, to drug discovery. All chapters were written by internationally leading researchers and clinicians in the field. It is our wish that readers discover the importance of polymyxin structure in relation to the mechanisms of activity, resistance and toxicity. We emphasized that reliable analytic methods for polymyxins are critical when investigating their pharmacokinetics (PK) and pharmacodynamics (PD). The complicated dose definitions and different pharmacopoeial standards have already compromised the safe use of polymyxins in patients. Therefore, informed by the latest pharmacological information, scientifically-based dosing recommendations have been proposed for intravenous polymyxins. Considering the PK/PD limitations and potential development of resistance, polymyxin combinations are encouraged; however, the current literature has not shown definite microbiological benefits, possibly because most clinical studies to date overlooked key PK/PD principles. Nephrotoxicity is the major dose-limiting factor and it is imperative to elucidate the mechanisms and develop novel approaches to minimize polymyxin-associated toxicities. In addition, the anti-endotoxin effect of polymyxins supports their clinical use to treat Gram-negative sepsis. Fortunately, the discovery of new-generation polymyxins with wider therapeutic windows has benefited from the latest achievements in polymyxin research. This book provides extensive pharmacological information on polymyxins to infectious diseases clinicians, pharmacists, clinical microbiologists, antimicrobial pharmacologists, and pharmaceutical scientists, and is an essential read for those who aim to develop novel polymyxins and improve their clinical use as a last-line defense against Gram-negative 'superbugs'.

This edited book explores the use of technology to enable us to visualise the life sciences in a more meaningful and engaging way. It will enable those interested in visualisation techniques to gain a better understanding of the applications that can be used in visualisation, imaging and analysis, education, engagement and training. The reader will be able to explore the utilisation of technologies from a number of fields to enable an engaging and meaningful visual representation of the biomedical sciences. This use of technology-enhanced learning will be of benefit for the learner, trainer and faculty, in patient care and the wider field of education and engagement. This second volume on Biomedical Visualisation will explore the use of a variety of visualisation techniques to enhance our understanding of how to visualise the body, its processes and apply it to a real world context. It is divided into three broad categories - Education; Craniofacial Anatomy and Applications and finally Visual Perception and Data Visualization. In the first four chapters, it provides a detailed account of the history of the development of 3D resources for visualisation. Following on from this will be three major case studies which examine a variety of educational perspectives in the creation of resources. One centres around neuropsychiatric education, one is based on gaming technology and its application in a university biology curriculum, and the last of these chapters examines how ultrasound can be used in the modern day anatomical curriculum. The next three chapters focus on a complex area of anatomy, and helps to create an engaging resource of materials focussed on craniofacial anatomy and applications. The first of these chapters examines how skulls can be digitised in the creation of an educational and training package, with excellent hints and tips. The second of these chapters has a real-world application related to forensic anatomy which examines skulls and soft tissue landmarks in the creation of a database for Cretan skulls, comparing it to international populations. The last three chapters present technical perspetives on visual perception and visualisation. By detailing visual perception, visual analytics and examination of multi-modal, multi-parametric data, these chapters help to understand the true scientific meaning of visualisation. The work presented here can be accessed by a wide range of users from faculty and students involved in the design and development of these processes, to those developing tools and techniques to enable visualisation in the sciences.

This book provides a cutting-edge review of polyglutamine disorders. It primarily focuses on two main aspects: (1) the mechanisms underlying the pathologies' development and progression, and (2) the therapeutic strategies that are currently being explored to stop or delay disease progression. Polyglutamine (polyQ) disorders are a group of inherited neurodegenerative diseases with a fatal outcome that are caused by an abnormal expansion of a coding trinucleotide repeat (CAG), which is then translated in an abnormal protein with an elongated glutamine tract (Q). To date, nine polyQ disorders have been identified and described: dentatorubral-pallidoluysian atrophy (DRPLA); Huntington's disease (HD); spinal-bulbar muscular atrophy (SBMA); and six spinocerebellar ataxias (SCA 1, 2, 3, 6, 7, and 17).The genetic basis of polyQ disorders is well established and described, and despite important advances that have opened up the possibility of generating genetic models of the disease, the mechanisms that cause neuronal degeneration are still largely unknown and there is currently no treatment available for these disorders. Further, it is believed that the different polyQ may share some mechanisms and pathways contributing to neurodegeneration and disease progression.
