Über DNA Barcoding: Philippine Cinnamomum Schaeffer (Lauraceae)
Philippine Cinnamomum Schaeffer family Lauraceae are trees with aromatic leaves and bark.It has several economic and medicinal uses, but concerns on its DNA barcodes are limited thus, this study was conducted.The 27 collected species were subjected to DNA isolation,Polymerase Chain Reaction and DNA Sequencing.A modified CTAB successfully isolated the DNA of the species.Amplification with rbcL and master mix provided 36 good DNA from 45 samples. BLAST results were 86% to 100% maximum identity.MEGA 7 analysis resulted to Cluster 1 composed of 34 individuals in 22 species with 75% bootstrap support. Four subclades were formed in the major clades and fourteen individual plant stood alone as singleton.Cluster 2 composed of two species as two singletons.Bootstrap branch are all above 50%, lowest 60% and highest of 75 %.Some of the species showed genetic divergence.The 36 DNA samples belong to only 14 haplotypes in 140 nucleotides sites.Haplotype 1 has 18 samples composed of 12 species.It confirmed that all individuals are Cinnamomum species. Similar species located in different geographic sites has slight difference in DNA sequences.
